TEM sample preparation for biological or soft materials (by staining and/or resin embedding) Biological or soft materials imaged under TEM tend to suffer from a lack of contrast (due to the low atomic weight of their elemental composition). Staining adds electron-dense material to the sample to improve contrast. Larger samples can also be stained and embedded in resin prior to further TEM sample preparation by ultramicrotomy. This short course teaches fundamentals of negative staining and/or resin embedding.
